Undesired protein phosphorylation by host kinases or autophosphorylation often complicates protein expression in standard bacterial strains. The BL21(λDE3) pLPP E. coli strain is specifically engineered for high-efficiency, homogeneous expression of non-phosphorylated proteins—including challenging serine/threonine kinases and tyrosine kinases.
Amid Biosciences' BL21(λDE3) pLPP chemically competent cells contain an IPTG-inducible bacteriophage lambda protein phosphatase (lambda PP) expression plasmid. Lambda PP is a highly specific enzyme that efficiently removes phosphorylation from Ser, Thr, and Tyr residues in target proteins during co-expression.
